Mechanisms underlying neurotransmission and neurodegeneration.

Neurotransmitters are released by exocytosis. We have studied the functions of mammalian proteins in exocytosis including Munc18-1 and cysteine string protein (Csp). Current work is focused on the nematode worm Caenorhabditis elegans as a model for the functional study of UNC-18 (orthologue of Munc18-1). We are also exploring the role of synaptic proteins in the protection of neurons from age-related neurodegeneration in the nervous system



Functions of calcium sensor proteins

Many aspects of cell function are regulated by cytosolic calcium acting through calcium-binding proteins. We are interested in two such families, the NCS proteins and the CaBPs. We are investigating the physiological roles of NCS-1 and related family members including the KChIPs which regulate the traffic of Kv4 potassium channels. Biochemical and structural approaches including use of NMR are used to identify interacting proteins and the molecular basis by which they act.

The localisation of the neuronal calcium sensor proteins proteins is analysed using fluorescent-tagged proteins and their calcium-myristoyl switch mechansim visualised in live cells.
